吲哚-3-甲醇通过抑制Survivin表达诱导人鼻咽癌细胞的凋亡 被引量:2

The pilot study on the mechanism of CNE1 cells apoptosis induced by I3C

摘要 目的探讨吲哚-3-甲醇在体外实验条件下诱导人鼻咽癌CNE1细胞增殖的影响及其诱导CNE1细胞凋亡的作用与可能机制。方法水溶性四氮唑(WST-1)法检测吲哚-3-甲醇在体外对人鼻咽癌CNE1细胞增殖的抑制作用;DAPI荧光染色法直接观察及通过检测线粒体膜电位反映吲哚-3-甲醇诱导的CNE1细胞凋亡现象;蛋白免疫印迹(Westernblotting)方法检测凋亡相关信号分子存活素(survivin)蛋白表达情况。结果吲哚-3-甲醇在体外可以剂量依赖的方式抑制人鼻咽癌细胞增殖,并可以诱导细胞凋亡,且在凋亡过程中,凋亡抑制蛋白survivin表达降低。结论吲哚-3-甲醇在体外可抑制人鼻咽癌CNE1细胞增殖并诱导其发生凋亡,且凋亡的发生可能与survivin表达受抑有关。 Objective To study the effect of indoel-3-carbinol on CNE1 cells as well as the mechanism underlying it.Methods The inhibition of cell proliferation was assay by WST-1 agent,and the apoptosis was shown with DAPI and JC-1 staining.To study the mechanism of apoptosis,the expressions of the survivin protein were detected with western immublotting assay.Results The results showed that I3C could inhibit the proliferation of CNE1 cells with the dose-dependent manner and the apoptosis could be induced,too.Further more,the expressions of survivin appeared to be decreased significantly as compared with those of the control.Conclusion It is suggested that I3C could induce apoptosis of CNE1 cells in the manner of dose-dependent,and survivin might play a role during above processes.
