摘要
目的研究黄体酮对实验性变应性脑脊髓炎(experimental allergic encephalomyelitis,EAE)大鼠的治疗作用。方法采用注射豚鼠脊髓匀浆的方法制作雄性Wistar大鼠EAE模型。当EAE大鼠出现神经症状后,开始在治疗组腹腔内注射溶于2.5ml芝麻油中的黄体酮(4mg·kg-1·d-1,连续注射4d),模型组注射等量的芝麻油。分别对两组大鼠进行运动功能评分和脑组织的电镜检查、LFB染色、锇酸Marchi's染色。染色结果采用IPP图像分析软件计算光密度(OD)值。结果治疗组和模型组神经功能评分无明显区别,但是在开始治疗后第6、10、25天,治疗组正常髓鞘的积分光密度值明显高于模型组,而退变髓鞘的积分光密度值明显低于模型组。结论黄体酮有利于EAE大鼠中枢神经系统的髓鞘再生,促进髓鞘修复。
Objective To study the therapeutic effect of progesterone for rats with experimental allergic encephalomyelitis(EAE).Methods The male Wistar rats were immunized with guinea pig s spinal cord homogenate.After neurological symptoms occurred in the rats,intraperitoneal injection of progesterone which was dissolved in 2.5 ml sesame oil was given to the therapeutic group at the dose of 4 mg/kg once a day for 4 d and the control group was given the same amount of sesame oil.All the rats were graded with the nerve function score. Their brains were examined by electron microscopy and stained by LFB and Marchi method. The IPP system was used to calculate the IOD values of normal and degenerative myelin. Results There was no signifi- cant difference in the nerve function score between therapeutic and control group. But in the 6th, 10th,25th day after progesterone injection, the IOD values of normal myelin of therapeutic group were higher than those of control group. The IOD values of degenerative myelin of therapeutic group were lower than those of control group. Conclusion Progesterone is profit to the remyelination of central nervous system of the rats with experimental allergic encephalomyelitis.
