家猫多焦视网膜电图、图形视网膜电图及图形视觉诱发电位的可重复性研究

摘要 目的探讨家猫图形视网膜电图(pattern electroretinogram,PERG)及图形视觉诱发电位(pattern visual evoked potential,PVEP)及多焦视网膜电图(multi-focal electroretino-gram,mfERG)检测的可重复性及其影响因素。方法健康成年家猫10只20眼,参照人类电生理记录的标准化方案及小鼠与大鼠标准化方案建议进行参数设置,以自行设计的定位装置进行了动物固定,共检测2次,每次检测均固定在相同的时间段,2次检测相隔1周,主要分析前后2次检测的可重复性及变异系数。结果mfERG检测:N1波5环的2次检测潜伏期分别为(15.01±6.61)ms、(14.86±6.49)ms,振幅密度分别为(6.54±3.19)nV.deg-2、(4.05±3.15)nV.deg-2,2次结果比较差异均有统计学意义(P均<0.05),其他指标2次检测有良好的重复性,结果比较差异均无统计学意义(P均>0.05)。变异性方面,P1波的潜伏期或振幅密度变异系数要较N1波小;振幅密度与潜伏期相比,潜伏期的变异系数小于振幅密度;N1波5个环的振幅密度与潜伏期随着离心度的增加,变异系数增大。家猫PERG之P50波、N95波,PVEP之P100波振幅和潜伏期稳定性及可重复性好,2次检测的差异均无统计学意义(P均>0.05)。以上波形潜伏期的变异系数较振幅小。可重复性检测的主要影响因素有固视、瞳孔大小、记录电极的位置、2次记录的间隔时间。结论家猫mfERG、PERG及PVEP具有较好的可重复性。记录参数及刺激参数相对固定时,固视、瞳孔大小、记录电极的位置(PVEP的记录)、2次记录的间隔时间等因素对家猫mfERG、PERG及PVEP检测结果的重复性具有明显的影响。 Objective To explore the repeatability and influencing factors of pattern electroretinogram(PERG),pattern visual evoked potential(PVEP) and multi-focal electroretinogram(mfERG) in cats.Methods The parameters were set at 10 healthy adult cats(20 eyes) with reference to standards of electrophysiology record in human and the proposed standards in mice and rats,and fixed with self-designed device.All cats were tested for twice,the two tests were with 1 week interval,but both were at the same time point.The repeatability and coefficient of variation were mainly analyzed between the two tests.Results The results of two tests showed that the peak latencies of N1 wave at 5 ring were(15.01±6.61)ms,(14.86±6.49)ms and the amplitude densities were(6.54±3.19)nV·deg-2,(4.05±3.15)nV·deg-2 by mfERG.There were statistical differences between two tests(both P〈0.05).There was good repeatability in other index,and no significant difference was found between the two tests(all P〉0.05).About coefficient of variation,incubation period and amplitude density of P1 wave were lower than those of N1 wave,incubation period was lower than that of amplitude density,and incubation period and amplitude density at 5 rings of N1 wave raised with the increase of centrifugal degree.Incubation period and amplitude of P50,N95 and P100 wave at PERG had good stability and repeatability,and no statistical difference was found between the two tests(all P〉0.05),but the coefficient of variation of incubation period of those waves were lower than those of amplitude.Key factors influencing repeatability included fixed vision,size of pupil,location of record electrode and intervals between two records.Conclusions The mfERG,PERG and PVEP show good repeatability in cats.Factors as fixed vision,size of pupil,location of record electrode and intervals between two records have significant influences on mfERG,PERG and PVEP in cats when parameters of record and stimulation are relatively unchanged.
