仙人掌对体外培养成纤维细胞生长增殖的影响

摘要 目的:观察仙人掌对体外培养成纤维细胞增殖的影响及其量-效关系。方法:以小鼠成纤维细胞株(NIH3T3)为研究对象,采用四氮唑盐(MTT)比色法测定仙人掌对成纤维细胞增生的影响(以OD值反映细胞活性),采用免疫细胞化学染色法检测成纤维细胞的增殖细胞核抗原(PCNA)的表达。结果:在一定浓度范围内(0.0625~0.5mg/mL),仙人掌能明显促进体外培养的成纤维细胞生长增殖,同时促进PCNA蛋白的表达,并随着仙人掌浓度的增加,成纤维细胞数量和PCNA蛋白的表达均不断增加,在0.5mg/mL浓度时达到高峰,呈现明显的剂量-效应关系(r分别为0.761,0.783;P均<0.01)。结论:在一定浓度范围内,仙人掌具有促进体外培养成纤维细胞增殖的作用,并表现为明显的量-效关系。 Objective: To explore the effects of Opuntia on fibroblast proliferation in vitro and its concentration-effect relationship. Methods: The effects of Opuntia on proliferation of fibroblast were measured with MTT colorimetric assay,and PCNA productions were examined with immunocytechemistry. Results: Opuntia could provoke fibroblast proliferation and increase PCNA protein expression in concen- tration range 0.0625~0.5 mg/mL. Furthermore,the effects exhibited a significant concentration-dependent relationship(r=0.761,0.783 respectively,P〈0.01). Conclusion: Opuntia can provoke fibroblast prolif- eration within a certain concentration range, which exhibits a significant concentration-effect relationship.
