摘要
目的探讨山楂叶总黄酮(FMCL)对脑缺血-再灌注(CIR)损伤的保护作用及其可能机制。方法采用线栓法制备大鼠局灶性脑缺血-再灌注损伤模型,对其进行行为指标评分,TTC染色测定梗死范围,HE染色检测脑组织病理变化,透射电镜检测神经元超微结构变化,并检测大鼠脑组织匀浆中超氧化物歧化酶(SOD)活性、丙二醛(MDA)及一氧化氮(NO)含量的变化。结果FMCL可明显改善大鼠神经行为,缩小梗死范围,减轻脑组织病理改变,并可提高脑组织中SOD活性,降低脑组织中MDA及NO含量(P<0.05或P<0.01)。结论FMCL可减轻CIR神经细胞损伤,其机制可能与抗自由基损伤、减轻NO神经毒性有关。
Objective To investigate the protective effects of flavone mixture of crataegus leaves(FMCL) on cerebral ischemia-reper- fusion(CIR) injury in rats. Methods The CIR model was built through thread block. The neurologie deficit score and infarction area were measured. Then pathological change in injury brain tissue was detected by HE stain and transmission electron microsc.ope. At the same time, the levels of superoxide dismutase (SOD) ,malondialdehyde (MDA) and nitric oxide(NO) in brain tissue were observed. Results FMCL reduced cerebral infarction area, and relieved the injury of brain tissue. It also increased the activity of SOD and decreased the contents of MDA and NO in a dose-dependent manner( P 〈 0. 05 or 0. 01 ). Conclusions FMCL has a protective effect on CIR injury. Its mechanism maybe relate to anti-radicals damage and lessen the neurotoxicity of NO.
