摘要
目的:本实验从mRNA水平研究肾癌组织、癌旁肾组织及正常肾组织中受体酪氨酸激酶EphB4和基质金属蛋白酶-9(MMP-9)的表达规律及二者表达之间的相关性,探讨其在肾癌发生发展过程中的可能作用,为肾癌的诊断、治疗及预后判断提供新的方法和思路。方法:运用半定量RT-PCR技术检测肾癌组织、癌旁肾组织及正常肾组织中EphB4和MMP-9在mRNA水平的表达情况,分析其与肾癌临床病理因素的相关性,探讨其与肾癌细胞增殖、浸润和转移的关系。结果:EphB4和MMP-9在肾癌组织、癌旁肾组织中都表达,且癌旁表达低于肿瘤组织表达,差异有统计学意义(P<0.05)。二者在正常肾组织中几乎不表达(P<0.01)。两者在肾癌组织中的表达丰度与患者的年龄、性别无关(P>0.05),与肿瘤的临床分期、病理分级以及是否淋巴结转移密切相关(P<0.05)。二者在肾癌组织中表达呈明显正相关关系(r=0.631,P<0.01)。结论:EphB4和MMP-9在肾癌的发生、发展及侵袭转移中起非常重要作用,联合检测两者的表达对预测肾癌患者预后、指导治疗有一定临床意义。
Objective:To study the expression of EphB4 receptor tyrosine kinase in renal cancer,corresponding adjacent renal tissue and normal renal tissue,to explore its relationship with matrix metalloproteinase-9(MMP-9) expression of renal cancer in mRNA level,and to investigate the possible role of EphB4 in the renal carcinogenesis for the diagnosis,therapy,and prognosis assessment. Methods:The expression of EphB4 and MMP-9 in mRNA level was detected by semiquantitative reverse transcriptase polymerase chain reaction in specimens of renal cancer,corresponding adjacent renal tissue and normal renal tissue,and the relationships between the expression of EphB4/MMP-9 and the clinicopathologic parameters were further investigated. Results:EphB4 and MMP-9 genes were expressed in both human renal cancer tissue and the corresponding adjacent tissue,and the expression in renal carcinoma tissue was more than in the corresponding adjacent tissue (P〈0.05). The normal renal tissue had low expression of EphB4 and MMP-9 genes (P〈0.01). Besides,high expression of EphB4 mRNA and MMP-9 mRNA was associated with pTNM stage and histological differentiation respectively (both P〈0.05),but not associated with age and gender (P〉0.05). There was a significant positive correlation between the expression of EphB4 mRNA and MMP-9 mRNA in renal carcinomas (r=0.631,P〈0.01). Conclusion:EphB4 plays a very important role in the carcinogenesis,invasion and metastasis of renal cell carcinoma. The combined expression of the two genes has certain significances in predicting the prognosis of patients with renal cell carcinoma and guiding the treatment.
