抗心磷脂抗体阳性脑梗死与HLA-DQA1和HLA-DQB1基因多态性的关系

The Relationship between Anticardiolipin Antibody and HLA-DQA1 and HLA-DQB1 Gene Polymorphism in Patients with Cerebral Infarction

目的:探讨人类白细胞抗原(HLA)的DQA1和DQB1基因多态性与抗心磷脂抗体(ACA)阳性脑梗死的相关关系。方法:运用聚合酶链式反应-序列特异性引物(PCR-SSP)法对30例抗心磷脂抗体阳性(ACA+)脑梗死患者和90例抗心磷脂抗体阴性(ACA-)脑梗死患者HLA-DQA1和DQB1基因多态性进行分型。结果:2组共检出12对HLA-DQA1和HLA-DQB1等位基因,ACA+脑梗死组患者的DQA1*0301等位基因频率为35.0%,高于ACA-脑梗死组患者的17.7%(P<0.05);ACA+脑梗死组患者的DQB1*0501等位基因频率为31.7%,高于ACA-脑梗死组患者的16.7%(P<0.05)。结论:DQA1*0301和DQB1*0501等位基因与ACA+脑梗死的易感性相关,可能是ACA+患者脑梗死的易感基因。

Objective： To investigate the relationship between HLA-DQAI and HLA-DQB1 gene polymorphism and anticardiolipin antibody in patients with cerebral infarction. Methods： Thirty patients of cerebral infarction with anticardiolipin antibody, 90 patients without anticardiolipin antibody were investigated for HLA- DQA1 and HLA-QDBI genotyping by polymerase chain reaction-sequence specific primers （PCR-SSP） in Tianjin of north China. Results： The frequency of HEA- DQAI＊0301 was 35% or 17.7% in patients of cerebral infarction with or without anticardiolipin antibody. The frequency of HLA-DQBI＊0501 was 31.2% or 16.7% in patients of cerebral infarction with or without anticardiolipin antibody. The difference of the frequencies of HLA-DQAI＊0301 and HLA-DQBI＊0501 was statistical significance in two groups of patients （P〈 0.05）. Conclusion： HLA-DQA1 ＊0301 and HLA-DQB 1＊0501 allele was positively related to the genetic susceptibility in patients of cerebral infarction with anticardiolipin antibody.
