真空水解测定鱼皮胶原蛋白制品中的羟脯氨酸 被引量:3

摘要 采用0.75%氯化亚锡、6M盐酸于真空安瓿中115℃、24h水解鱼皮胶原蛋白制品中的羟脯氨酸,用对二甲胺基苯甲醛显色,560nm比色测定羟脯氨酸含量,测定结果鱼皮胶原蛋白寡肽的羟脯氨酸含量为2.57%,鱼皮胶原蛋白颗粒的羟脯氨酸含量为1.50%,回收率97.8%-103.2%,真空水解方法简便,测定结果稳定,重复性好,一次可水解批量样品,提高工作效率。 The collagen of fish skin was hydrolyzed under the following conditions: temperature of 115 ℃, vacuum SnCl2 enncentration of 0.75%, HCl concentration of 6 mol/L and hydrolyses time of 24 hours. The hydroxyproline was determined by speclmphotometry at 560 nm with paradimethylaminobenzal as colourant. Results showed that the content of hydroxyproline in the collagen oligopeptide was 2.75%, and the content of hydroxyproline in the collagen was 1.50%. And recovery of the method was in range of 97.8 %- 103.2 %.
