摘要
利用大花萱草的茎尖离体诱导愈伤组织,先筛选出大花萱草组培离体再生系统,在该培养基中添加不同浓度NaCl做胁迫条件,统计组培苗的生长情况,并测定了组培苗的脯氨酸含量的变化。试验表明:金娃娃和江苏沭阳萱草在MS+NAA 0.4 mg/L+6-BA 4.0 mg/L培养基上的诱导愈伤率较高。组培苗叶片生长量随盐浓度的增加而递减,主要表现为叶片的生长受到抑制,随着盐浓度的增高脯氨酸的含量表现越高,相同NaCl胁迫浓度下江苏沭阳萱草脯氨酸含量高于金娃娃。
The calli were induced by shoot tip of Hemerocallis middendorfii.The results showed that rate of inducing calli of Hemerocallis middendorfii were high by using medium of MS+NAA 0.4 mg/L+6-BA 4.0 mg/L.The growth of tissue culture plant were abserved and proline of tissue culture plantwere determined on the medium added different concentration Sodium chloride.With concentration of Sodium chloride increasing.The experiment showed that the growth rate of tissue culture plant declined, growth of leave were inhibited and the content of proline were increasing. The contenl of proline of Jiangsu Shuyang was higher that of Hemerocallis fulva var. flore-pleno at same concentration of Sodium chloride.
