摘要
目的观察GnRH-A制剂——抑那通(注射用醋酸亮丙瑞林)诱导的特发性性早熟小鼠动物模型与阴虚火旺证的相关关系及知柏地黄丸的干预作用。方法采用抑那通诱导NIH雌性小鼠建立特发性性早熟模型,观察该模型的体温、体重、血清环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)、cAMP/cGMP的变化;以甲地孕酮为阳性对照药,观察知柏地黄丸对上述指标变化的影响。结果抑那通诱导的模型动物表现出体质量及卵巢重量增加,子宫重量亦有增加的趋势;高、低剂量的知柏地黄丸及甲地孕酮均能对抗抑那通所致动物体温升高和卵巢重量的增加;低剂量的知柏地黄丸及甲地孕酮均能对抗抑那通所致动物血清cAMP含量升高。此外,除正常对照组外,各组血清cAMP含量与体温及卵巢重量之间均呈正的直线相关;阴虚火旺证模型组动物血清cAMP含量与子宫重量之间亦呈正的直线相关。结论抑那通诱导的特发性性早熟模型同时表现出阴虚火旺证的特征表现。知柏地黄丸在抑制抑那通诱导的性成熟提前的同时,亦能拮抗其诱导的阴虚火旺证的特征表现。表明抑那通诱导的特发性性早熟模型与阴虚火旺证之间具有一定的相关性。
Objective To observe the correlation between idiopathic central precocious puberty(ICPP) in mice induced by gonadotrophin releasing hormone agent( Gn- RH-A), Leuprorelin, and the syndrome of yin-deficiency and fire-hyperactivity, as well as the intervention effect of "Zhibo Dihuang Pill" on it. Methods ICPP mice model was established by Leuprorelin. The effect of "Zhibo Dihuang Pill" on the body temperatures( BTs), body weights( BWs), the levels of the cAMP, cGMP and cAMP/eGMP in serum were observed, and Megetrol acted as control drug. Results The BWs and the weights of ovaries of ICPP models increased, but were inhibited by "Zhibo Dihuang Pill" and Megetrol. The elevated levels of the cAMP in serum of ICPP models were depressed by low-dose "Zhibo Dihuang Pill" and Megetrol. The levels of the cAMP in serum, the BTs, and the weights of ovaries showed linear correlation except normal control group. The levels of the cAMP in serum and the weights of uterus of the group of yin-deficieney and fire-hyperactivity syndrome showed linear correlation too. Conclusion The ICPP mice model induced by Leuprorelin displays the characteristics of yin-deficiency and fire-hyperactivity syndrome. "Zhibo Dihuang Pill" can inhibit the precocious puberty, meanwhile antagonizes the characteristic manifestation of yin-deficiency and fire-hyperactivity syndrome. The results indicate that a certain correlationship exists between the ICPP mice model induced by Leuprorelin and yin-deficiency and fire-hyperactivity syndrome.
