摘要 目的初步探讨永生化人脐静脉内皮细胞(Human umbilical vein endothelial cells,HUVEC)构建血管瘤疾病模型的可行性。方法永生化HUVEC与猪骨髓间充质干细胞(BMSC)分别在普通的DMEM培养液(含10%FBS)中贴壁培养至完全融合后,胰酶消化为单细胞悬液;细胞计数后按不同细胞组合分组,与Matrigel混合后分别注射至裸鼠皮下,移植后1周及5周取材,HE染色观察血管生成情况。结果1周后HE所见:空白组与单纯注射BMSC组均无管样结构形成;单纯注射HUVEC组和注射混合细胞组均有管样结构形成。5周后HE所见:空白组被吸收;单纯注射BMSC组可见大量的脂肪组织;注射单纯HUVEC组以及混合组可见血管样结构存在。结论HUVEC在裸鼠皮下注射具有形成血管的能力,有望构建增生期血管瘤模型。 Objective To investigate the feasibility for hemangioma model by using immortalized human umbilical vein endothelial cells (HUVEC) constructing. Methods HUVEC and pig bone marrow stromal cells (BMSC) were cultured in regular DMEM with 10% fetal bovine serum. Cells were trypsinized, counted, and re-suspended in Matrigel. The cell/gel mixtures were then injected subcutaneously into the back of 6-week-old nude mice. The grafts were harvested after 1 week and 5 weeks for histological analysis. Results After 1 week, no vascular structure was observed in the groups injected with gel or with BMSC alone. On the contrary, cord-like structures were observed in the groups injected with HUVEC alone or with HUVEC/BMSC mixture. After 5 weeks, many vascular structures were observed in both HUVEC group and HUVEC / BMSC group, while no tissue was found in the group injected with gel alone and only adipose tissue was observed in the group injected with BMSC alone. Conclusion HUVEC could form vascular structure in vivo, indicating that it might be an model for infantile hemaugioma in vivo.
