摘要
目的采用液相芯片技术分析上海地区尖锐湿疣患者人乳头瘤病毒(HPV)型别的分布情况。方法从116例尖锐湿疣患者的疣体标本中提取脱氧核糖核酸(DNA),应用液相芯片技术进行HPV基因型检测,并与聚合酶链反应(PCR)的检测结果进行比较,明确液相芯片的检测效能。结果液相芯片检测的116例标本中,HPV阳性114例,阳性率为98.3%。其中单一型别感染占71.6%(83/116),双重型别感染占18.1%(21/116),三重型别感染占7.8%(9/116),四重型别感染占0.9%(1/116)。共检出18种HPV亚型,其中HPV6和HPV11最为常见,感染率分别为60.3%(70/116)和42.2%(49/116)。在高危型HPV感染中,最常见的3种亚型依次为HPV16、HPV52和HPV18。液相芯片与PCR检测结果的符合率为90.7%,并且液相芯片对多重感染的检出率高于PCR。结论上海地区尖锐湿疣以HPV6和HPV11感染为主。高危型HPV感染中最常见的3种亚型依次为HPV16、HPV52和HPV18。液相芯片对HPV多重感染的检出率高于PCR。
Objective To use liquid chips (luminex method) for investigating the distribution of human papillomavirus (HPV) genotypes in patients with condyloma acuminata in Shanghai. Methods The biopsy specimens were obtained from 116 patients with condyloma acuminate and the deoxyribonucleic acids were extracted for analysis using liquid chips; the results were compared with those obtained by type-specific polymerase chain reaction (PCR) so as to assess the efficacy of liquid chips. Results Liquid chips showed that 114 of the 116 specimens were HPV positive, with the positive rate being 98.3%, including 71.6% (83/116) with single infection, 18.1% (21/116) with double infection, 7.8% (9/116) with triple infection and 0.9% (1/116) with quadruple infection. Eighteen genotypes were identified by liquid chips, among which HPV 6 and HPV 11 were the two most common types, with infection rates being 60.3% (70/116) and 42.2% (49/116), respectively. The top three high-risk HPV genetypes were HPV16, HPV52 and HPV18. The consistent rate between liquid chips and PCR for HPV genotyping was 90.7%. Besides, liquid chip had a high sensitivity than PCR for detection of multiple infection. Conclusion HPV6 and HPV11 are the 2 major HPV genotypes in patients with condyloma acuminata in Shanghai. The top three high-risk HPV genotypes are HPV16, HPV52 and HPV18 in order. The liquid chips have a higher sensitivity than PCR for dectection of HPV. (Shanghai Med J, 2009, 32, 892-895)
