摘要
目的了解NF-κB在子宫内膜异位症发病机制中的作用。方法分别用LPS及LPS+PDTC干预正常及内异症患者的在位及异位子宫内膜细胞,收集细胞后用WestBlot方法检测并比较三组NF-κB的蛋白浓度变化。结果(1)在正常、在位及异位子宫内膜细胞这三组细胞中,NF-κB蛋白在正常子宫内膜细胞表达最弱,与在位及异位子宫内膜细胞相比,差异有显著统计学意义(P<0.01);其次是在位子宫内膜细胞,异位子宫内膜细胞NF-κB的蛋白表达最高,两组之间比较,其差异有显著统计学意义(P<0.01)。(2)对在位及异位子宫内膜细胞给予LPS刺激后,NF-κB的蛋白表达明显增强,与对照组相比,差异有显著统计学意义(P<0.01);但如果在给予LPS刺激前先给予PDTC,则可见NF-κB的蛋白表达明显降低,与LPS组相比,差异有显著统计学意义(P<0.01)。LPS+PDTC组的NF-kB的蛋白表达甚至显著低于对照组,差异有显著统计学意义(P<0.01)。结论NF-κB可能在子宫内膜异位症的发病机制中起重要作用。
Objective To obtain the role about NF - κB in pathogenesis of endometriosis. Methods We used LPS and LPS + PDTC to intervene normal,eutopic and ectopic endometriotic cells. After collecting cells,we detected the density of NF - κB with West- Blot. Results ( 1 ) Comparing with the eutopic and ectopic endometriotic cells, protein NF - κB was the lest expression in the endometrial cells. The difference had uncommonly statistical significance( P 〈 0. 01 ). (2) After giving LPS to eutopic and ectopic cells, the protein expression of NF - κB obviously became stronger. As compared with the control group, the difference had notable stastical significance (P 〈 0. 01 ) ; but if PDTC was added before LPS, the protein of NF - κB became obviously lower. As compare with group, the difference of LPS had notoble stastical significance (P 〈 0.01 ). The protein expression of NF - κB in group LPS + PDTC became lower than that in control group. The difference had notable statistical signifcance ( P 〈 O. 01 ). Conclusion NF - κB may play important rote in the pathogenesis of endometriosis.
