海马放射状胶质细胞体外诱导激活后的PEBP mRNA表达变化

摘要 目的:探讨切割穹窿海马伞侧海马提取液对海马放射状胶质细胞中磷脂酰乙醇胺结合蛋白(PEBP)mRNA表达的影响。方法:将培养的海马胶质细胞接种于培养板中,分成诱导组和对照组,诱导组中加入含5%切割穹窿海马伞侧海马提取液的DMEM/F12培养液,对照组加入单纯的细胞培养液。培养7d后,提取总RNA,然后逆转录合成cDNA,用real-timePCR观察诱导组及切割组中PEBPmRNA的表达情况。结果:PEBPmRNA在放射状胶质细胞中能够表达,诱导组的表达比对照组明显增加(P<0.05)。结论:表达增加的PEBPmRNA可能与切割侧海马提取液"激活"的放射状胶质细胞向胆碱能神经元分化有关。 Objective: To observe the phosphatidylethanolamine binding protein (PEBP) expression in radial glial cells which were induced by the extracts from hippocampus after fimbria-fornix transection. Methods: Hippocampal glial cells were plated into 24-well plates and then divided into the induced group and the control group. The DMEM/F12 medium containing 5% extracts from hippocampus after fimbria-fornix transection was added into the induced group, while the control group merely added to the euhure medium. After 7days, the total RNA was extracted and then the first strand cDNA was synthesized, the real-time PCR was used to observe the expression changes of PEBP mRNA in radial glial cells. Results: PEBP mRNA can express in radial glial cells, the expression of induced group was significantly higher than that in the control group. Conclusion: The higher PEBP mRNA expression in the radial glial cells which were activated by the extracts from fimbria-fornix transected hippocampus may be involved with the radial gtial cells differentiating into cholinergic neurons.
