佛手水煎剂对RAW264.7癌细胞增殖的影响

Effects of fingered citron decoction on proliferation of RAW 264.7 cells in vitro

为了观察佛手水煎剂对RAW 264.7癌细胞增殖的影响,用生药量0.625,1.250,2.500和5.000mg/mL的佛手水煎剂处理RAW 264.7癌细胞,采用四甲基偶氮唑蓝(MTT)法测定细胞活力、台盼蓝排斥法测定细胞存活率、吖啶橙/溴化乙锭(AO/EB)荧光染色法检测细胞凋亡与坏死.结果表明:与对照组比较,0.625,1.250和2.500 mg/mL佛手水煎剂处理后,RAW 264.7癌细胞固缩,细胞核染色质凝聚、片断化,有凋亡小体出现,表现出典型的细胞凋亡特征;5.000 mg/mL佛手水煎剂处理后,RAW 264.7癌细胞肿胀,细胞膜破裂,呈现坏死症状,RAW 264.7癌细胞的增殖明显受到抑制(P<0.01),半数抑制浓度(IC50)为2.073mg/mL.说明佛手水煎剂能诱导癌细胞凋亡,抑制癌细胞增殖.

It was observed the effect of fingered citron decoction on the proliferation of RAW 264.7 cells in vitro.RAW 264.7 cells were exposed to fingered citron decoction with concentration gradients of 0.625,1.250,2.500 and 5.000 mg/mL,MTT was employed to assay cell viability and typan blue exclusion method was used to assess the survival rate.Further,AO/EB double staining was applied to observe apoptosis or necrosis.The results showed that,compared with the control group,0.625~2.500 mg/mL of fingered citron decoction obviously induced apoptosis in RAW 264.7 cells,showed the apoptotic changes in morphology such as cell shrinkage,chromatin condensation and apoptotic bodies,in contrast,5.000 mg/mL of which induced necrosis in RAW 264.7 cells including cellular swelling and ruptured membranes.Decoction of fingered citron could significantly inhibit the viability of RAW 264.7 cells（P0.01）,the value of IC50 was 2.073 mg/mL in vitro.This suggested fingered citron decoction could effectively inhibit the proliferation of RAW 264.7 cells.
