摘要
【目的】观察丙泊酚对成年大鼠急性缺血缺氧后脑脊液(cerebrospinal fluid,CSF)内谷氨酸(Glutamate,Glu)、多巴胺(dopamine,DA)和乳酸脱氢酶(lactic acid dehydrogenase,LDH)浓度变化的影响。【方法】采用单侧颈总动脉结扎合并吸入氮气(N2)法制备大鼠急性缺血缺氧模型,随机分为实验组和对照组各10只,实验组给予丙泊酚5.0μg/g后再给予缺氧,对照组直接给予急性缺氧,在不同时间段检测CSF内Glu、DA和LDH的浓度变化。【结果】CSF内Glu、DA与LDH的基础值实验组与对照组相比无明显差异。急性缺氧后,Glu浓度在实验组与对照组均有上升趋势。实验组与对照组相比Glu水平升高减少(P<0.05)。DA浓度实验组30 min、60 min时与对照组相比升高明显减少(P<0.05),在120 min时两组无明显差别。LDH浓度实验组与对照组相比在120 min时升高减少(P<0.05)。【结论】急性缺血缺氧可使大鼠CSF内Glu、DA和LDH水平升高,丙泊酚可降低这些递质的升高幅度,具有脑保护作用。
【Objective】To observe the effect of propofol on changes of Glutamate(Glu),dopamine(DA) and lactic acid dehydrogenase(LDH) in cerebrospinal fluid(CSF)induced by acute hypoxia-ischemia in rats.【Methods】The rat model of acute hypoxia-ischemia was made by combining unilateral ligation of common carotid artery with inhaling nitrogen(N2).Rats were divided into two groups at random: control group and propofol group,and every group includes 10 rats.After all baseline concentrations were recorded,rats in propofol group were pretreated with propofol at 5.0 μg/g followed with inhaling N2 and rats in control group were given N2 directly.The changes of concentration of Glu,DA and LDH in CSF were detected at different time with HPLC.【Results】The baseline concentrations of Glu,DA and LDH had no significant changes in two groups(P〈0.05).After acute hypoxia,the concentrations of Glu increased in both groups,but concentrations in propofol group at 60 min and 120 min were significant lower as compared with those in control group(P〉0.05).The concentrations of DA in propofol group at 30 min and 60 min were significant lower than those in control group(P〈0.05),but concentrations at 120 min have no significant change in both groups.The increased concentration of LDH at 120 min after acute hypoxia in propofol groups was higher than that of control group(P〈0.05).【Conclusions】The concentrations of Glu,DA and LDH in CSF induced by acute hypoxia-ischemia increase and propofol may inhibit the increase with the effect of brain protection.
