摘要
目的:探讨胃泌素受体拮抗剂丙谷胺(PGL)对人结肠癌裸鼠移植瘤的作用。方法:建立人结肠癌SW480细胞株裸鼠移植瘤模型,观察在PGL、5肽胃泌素(PG)作用下移植瘤的体积和重量,用放免法测CAMP、流式细胞术测DNA、蛋白质和细胞周期。结果:移植瘤的体积和重量、瘤细胞内CAMP、DNA和蛋白质含量、S和G2M期细胞数及增殖指数PG组均显著高于对照组,PGL组与对照组比均无显著差异;PG+PGL组均显著低于PG组,而与对照组比均无显著性;G0/G1期细胞数PG组明显低于对照组,PGL组与对照组比无显著差异,PG+PGL组显著高于PG组,而与对照组比无显著性。结论:PGL对人结肠癌SW480细胞株移植瘤的生长无明显影响,但可抑制PG对移植瘤的促增殖作用,其作用是通过拮抗PG促进瘤细胞内CAMP、DNA和蛋白质的合成,抑制瘤细胞从G0/G1期进入到S。
Purpose: To explore the effect of proglumide (PGL), a gastrin receptor antagonist on the xenografted human colon cancer in nude mice. Methods: Establish the model of xenografted human SW480 cell line colon cancer in nude mice. Determine the volume and weight of xenografted cancer treated with PGL and pentagastrin (PG). Determine the content of cAMP by radioimmunoassy and the content of DNA, protein and cell cycle by flow cytometry. Results: The volume, weight, content of cAMP, DNA, protein, cell amount in S and G2M phase, cell proliferation index of xenografted cancer in PG group were markedly higher than those in control group, while those in PGL group did not differ from those in control group. Those in PG+PGL group were markedly lower than those in PG group, but did not differ from those in control group. Cell amount of G0/G1 phase in PG group was markedly lower than that in control group. In PGL group, it did not differ from that in control group. In PG+PGL group, it was markedly higher thah that in PG group, but did not differ from that in control group. Conclusion: PGL could not affect the growth of xenografted human SW480 cell line colon cancer, but could inhibit the growth promoting effect of PG on xenografted cancer through inhibiting cancer cell growing from G0/G1 to S, G2/M phase by antagonizing the promotng effect of gastrin on the synthsis of cAMP, DNA and protein in transplanted cancer cell.
