摘要
目的和方法:应用心脏微血管内皮细胞体外培养模型,研究不同浓度的TNFα、IL-1β、IL-6对微血管内皮与中性粒细胞粘附的调节。结果:5×104U/L的TNFα和IL-1β可明显增加内皮细胞与中性粒细胞的粘附(诱导内皮6h),并分别于105U/L、2×105U/L浓度时使中性粒细胞粘附数达高峰,但不同浓度的IL-6均未使粘附增加,免疫组化ABC法证实了同样的结果。进一步用抗ICAM-1和抗LFA-1单克隆抗体阻断实验表明,两种抗体在5mg/L浓度下可显著阻断中性粒细胞与内皮细胞间的粘附。结论:心脏微血管内皮细胞与中性粒细胞的粘附主要通过粘附分子ICAM-1/LFA-1介导。
Abstract AIM and METHOD:The effect of TNFα,IL-1β and IL-6 on the neutrophils-cardiac microvascular endothelial cells adhesion were investigated RESULTS:Adherence of rat neutropils to endothelial cells was significantly stimulated by TNFα(50U/mL)and the same consentration of IL-1β( P <0 05).The effect of TNFα and IL-1β tended to be dose dependent.On the other hand,IL-6 had no significant effect on adhesion of neutropils-endothelium.In the immunohistochemical study,cultured endothelial cells constitutively expressed ICAM-1,and ICAM-1 expression was significantly stimulated by TNFα and IL-1β. Meanwhile,adhesion of neutropils-endothelial cells stimulated by TNFα and IL-1β could be partly blocked by anti ICAM-1 and anti LFA-1 antibodies.The blockage rate was 56 2% and 47.3% respectively.CONCLUSION:Adherence of neutropils to endothelial cells was chiefly mediated by ICAM-1/LFA-1 MeSH Endothelium;Cells;Tumor necrosis factor;Interleukins;Neutrophils
