摘要
目的观察Tween-80合并温热对人膀胱癌阿霉素耐药细胞株的逆转作用及其机制。方法采用MTT法进行体外药物逆转实验,免疫细胞化学染色显示处理前后耐药细胞P-糖蛋白表达情况。结果①Tween-80合并41℃温热作用前后,EJ/ADM对抗癌药物阿霉素、丝裂霉素、5-氟尿嘧啶等药物的敏感性显著增加(P<0.01);②Tween-80合并41℃温热能够协同抑制p-gp蛋白的表达。结论Tween-80合并41℃温热对人膀胱癌阿霉素耐药细胞的耐药性有逆转作用,逆转机制与其抑制p-gp蛋白的表达有关。
Objective To investigate the reversal of drug resistance in the ADM-resistant human bladder cancer cell lines EJ/ADM by Tween-80 in combination with hyperthermia, and its mechanism. Method The reversing drug resistance was studied by MTT and the expression of P-glyeoprotein were examined by immunocytochemistry. Result MTT eytotoxic assay revealed that the sensitivities of EJ/ ADM cells to ADM, MMC, 5-FU, HCPT, VCR were different according to presence or absence of Tween80 combined with hyperthermia (41℃)(P〈0.01). The immuno-cytochemistry showed that the P- glycoprotein distributes in cellular membrane and cytoplasm, and after treated with Tween80 combined with hyperthermia (41℃), P-glycoprotein staining was redused in EJ/ADM cell. Conclusion The drug resistance of EJ/ADM cells could be reversed by Tween-80 in combination with hyperthermia(41℃).
