摘要
目的:研究抵抗素对人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)胰岛素信号通路的影响。方法:原代培养HUVECs,以不同水平人抵抗素(0~100μg/L)培养24h。用免疫印迹法检测其对经胰岛素作用的内皮细胞的磷酸化蛋白激酶B(phospho-protein kinase B,p-PKB)及磷酸化内皮型一氧化氮合酶(phospho-endothelial nitric oxide synthase,p-eNOS)水平,并对检测结果进行分析。结果:与无加抵抗素的对照组比较,抵抗素干预HUVECs 24h后,胰岛素刺激状态下的p-PKB和p-eNOS水平均明显降低(P〈0.05~0.01);基础状态下,抵抗素作用下HUVECs的p-PKB水平无明显改变(均为P〉0.05),而p-eNOS水平明显降低(P〈0.05):结论:抵抗素对内皮细胞的胰岛素信号转导和内皮型一氧化氮合酶磷酸化可起抑制作用,这可能是抵抗素诱导内皮细胞功能异常的机制之一。
Objective: To investigate the effects of resistin on insulin signaling in human umbilical vein endothelial cells(HUVECs). Methods: HUVECs were primary cultured, then treated with recombinant human resistin (0 - 100 μg/L) for 24 h. The levels of phospho-protein kinase B(p-PKB) and phospho-endothelial nitric oxide synthase(p-eNOS)were measured by western blotting in HUVECs under insulin stimulated conditions. Resuits: Compared with control group without resistin, the levels of p-PKB and p-eNOS in HUVECs by treatment 24 h with resistin were downregulated under insulin stimulated condition ( P 〈 0. 05 - 0. 01 ). Under basal condition, the level of p-PKB in HUVECS by treatment with resistin was not reduced, whereas it showed a decrease of p-eNOS ( P 〈 0. 05 ). Conclusion : Resistin exerts an inhibitory effect on the insulin signaling and eNOS phosphorylation in endothelial cells. It may be one of the mechanisms that resistin induces parafunction of endotheliocyte.
