Screening and analysis of biomarkers of esophageal cancer by combined technology of size exclusion chromatography and matrix assisted laser desorption ionization-time of flight-mass spectrometry

摘要 目的探索建立柱层析和基质辅助激光解吸电离飞行时间(MALD I-TOF)质谱技术筛选食道癌血清标志多肽的方法。方法选用Sephadex G-100葡聚糖作为分离介质,自制小型分离层析柱,分别采集30名健康人、30例食道良性疾病患者和30例食道癌患者血清进行多肽分离,结合MALD I-TOF质谱仪鉴定。结果通过多肽质荷比(m/z)比对技术,筛选出4种诊断食道癌疾病的潜在血清标志多肽,m/z分别为7 651.76、8 091.34、9 264.98、11 784.79。结论柱层析和质谱技术筛选食道癌血清标志多肽所建立的分析技术不仅快速简单,而且同样适合于筛选其他人类重大疾病的血清标志多肽或酶蛋白。 Objective To explore and set up a combined method of separation column of size exclusion chromatography (SEC) and matrix assisted laser desorption ionization-time of flight-mass spectrometry (MALDI-TOF-MS) for screening and analyzing the polypeptide and protein biomarkers of esophageal cancer. Methods The serum polypeptides in 30 healthy subjects, 30 benign esophageal disease patients and 30 esophageal cancer patients were pre-separated by the self-made separation column of Sephadex G-100, and the collected eluents were concentrated for MALDI-TOF-MS analysis respectively. Results With the technology of polypeptide mass change ratio comparison approach, four different ions m/z at 7 651.76, 8 091.34, 9 264.98, 11 784.79 were found, which were existing only in esophageal cancer patients. Conclusions The combined technology of separation column of SEC and MAIDI-TDF-MS is a simple and rapid method for screening esophageal cancer patients and it also can be applied to the rapid diagnosis of other diseases.
