中华鳖卵对高血糖模型大鼠血糖代谢的影响研究 被引量:3

摘要 目的研究中华鳖卵对高血糖模型大鼠糖脂代谢的影响。方法雄性SD大鼠以四氧嘧啶诱导高血糖模型,选择42只高血糖模型大鼠,按照体重和空腹血糖水平分为高血糖对照组(HC)和0.5、1.5和4.5 g/kg体重中华鳖卵冻干粉(TEP)实验组,另选8只健康雄性SD大鼠作为空白对照组(NC)。TEP实验组大鼠以灌胃方式给予不同剂量的TEP,HC和NC组给予等体积溶剂灌胃,实验周期8周。实验期间定期观察体重、进食量、饮水量和24 h尿量,实验结束时进行口服糖耐量实验(OGTT),检测空腹血糖、胰岛素水平,并采用免疫组织化学方法观察胰岛β细胞。结果中华鳖卵冻干粉可明显改善四氧嘧啶引起的进食量、饮水量、尿量增加和体重增长抑制,4.5g/kg体重TEP可促进高血糖模型大鼠的胰岛β细胞修复,升高空腹胰岛素水平,从而改善四氧嘧啶引起的口服糖耐量损伤,并降低空腹血糖。结论TEP对四氧嘧啶引起的血糖代谢紊乱具有明显的改善作用。 Objective To observe the effects of Chinese soft-shell turtle egg on blood glucose metabolism in alloxan-indueed hyperglycemic rats. Methods Male SD rats were injected (ip) with alloxan (200 mg/kg bw) to establish hyperglycemia model. Forty-two hyperglycemic rats were divided into four groups according to body weight and serum glucose level, namely hyperglycemia control (HC) group and 3 Chinese soft-shell turtle egg powder (TEP) treatment groups. Another 8 heahhy male SD rats were used as negative control (NC). Rats in TEP groups were treated by intragastric way with TEP at the dose of 0. 5, 1.5 and 4. 5 g/kg bw, respectively, for 8 weeks, while rats in HC and NC groups were treated with solvent in the same manner. The body weight, food intake, water intake, 24h urine output were recorded regularly during the experimental period. At the end of the treatment, an oral glucose tolerance test (OGTF) was carried out and the contents of fasting serum glucose (Glu), insulin (^125I radioimmuno-detection kit ) were tested in all groups. Pancreatic islet β cell restoration was also evaluated by immunohistoehemistry method in NC, HC and 4. 5 g/kg bw TEP group. Results Compared with HC group, TEP improved weight gain remarkably, but reduced water intake, food intake and 24 h urine output. A dose of 4. 5 g/kg bw TEP could improve β cell amount in pancreatic islet, thus increased fasting serum insulin level, reduced the 2 h serum glucose level and the area under glucose curve evidently in OGTT, and decreased fasting serum glucose level in alloxan-induced hyperglycemic rats. Condusion TEP is effective in improving glucose metabolic dysfunction in alloxan-indueed hyperglycemic rats.
