摘要
目的:研究心痹通(TKAM)对麻醉开胸犬冠脉结扎所致急性心肌缺血的保护作用,并观察其对相关血流动力学参数的影响。方法:犬冠状动脉结扎致心肌缺血模型,心痹通高(58.7 mg.kg-1)、中(29.3mg.kg-1)、低(14.7 mg.kg-1)剂量组十二指肠给药后采用心外膜电图标测心肌损伤范围及损伤程度,采用氯化硝基戊四氮唑兰(NBT)染色法测定心肌梗死区域。测定血清中肌酸激酶(CK)、乳酸脱氢酶(LDH)活力,以及丙二醛(MDA)含量。通过电磁血流量计探头观测心痹通对犬血流动力学参数的影响。结果:心痹通高、中剂量组可显著减轻心肌缺血程度并减小心肌梗死范围;心痹通高、中剂量组能明显降低血清CK和LDH活力,并降低血清MDA含量。心痹通高、中剂量组能显著改善麻醉犬相关血流动力学参数。结论:心痹通对犬冠脉结扎所致心肌缺血具有显著保护作用,并可明显改善相关血流动力学参数。
Objective:To study the effect of Trichosanthes kirilowii Allium macrostemon(TKAM) on acute myocardial ischemia induced by ligation of coronary artery,and on the hemodynamic parameters in anesthetized thoraco-opened dogs.Methods:Myocardial ischemia was developed by ligation of the left coronary anterior descending artery in the anaesthetized thoraco-opened dogs.TKAM at 58.7,29.3 and 14.7 mg·kg-1 was administrated.The epicardial electrocardiogram and the quantitative histological analysis combined with nitroblue tetrazolium (NBT) staining were used to determine the range and degree of the damage induced by myocardial ischemia. The activities of serum CK,LDH and MDA were determined. The effect of TKAM on hemodynamic parameters was ob- served by electromagnetic flowmetry. Results: TKAM significantly decreased the severity of myocardial ischemia and the extent of myocardial infarction. TKAM obviously reduced serum CK and LDH activities, and MDA content, and also improved the hemodynamic parameters. Conclusion:TKAM can protect anesthetized thoraco-opened dogs from myocardial ischemia induced by ligation of coronary artery; it can also improve the hemodynamic parameters.
