动脉粥样硬化兔自体血清在树突状细胞体外培养中的作用 被引量:2

摘要 目的:采取球囊损伤加高胆固醇饮食的方式建立兔动脉粥样硬化模型,观察模型兔自体血清对外周血源性树突状细胞的作用。方法:8只雄性新西兰白兔球囊损伤腹主动脉后给予高胆固醇饲料喂养8周,培养模型兔外周血源性的树突状细胞,随机分为4组,空白组加入PBS液,对照组及实验组分别加入制模前后两次提取的自体血清培养(10%),ox-LDL组加入ox-LDL(10μg/mL)。流式细胞学检测细胞表面标记物CD14、CD86、MHCⅡ的表达,观察不同浓度的同种异体混合淋巴细胞反应。结果:球囊损伤后高胆固醇饮食8周免动脉粥样硬化模型形成,实验组自体血清共培养后的DCs表面CD86和MHCⅡ的表达明显升高。并且由CD14^+转化为CD14^-,空白组及对照组DC表型无明显变化。且实验组DCs诱导的T淋巴细胞增值反应明显高于空白及对照组。ox-LDL组则产生和实验组同样的效应,且更加强烈。结论:通过球囊损伤加高胆固醇饮食的方式可以在短时间内建立兔动脉粥样硬化模型,模型兔体内存在诱导DCs成熟的抗原,DCs成熟与该动物模型AS发生发展可能有相关关系。
