乙肝患者肝组织和淋巴细胞内HBV检测及存在状态

THE PRESENCE OF HBV DNA AND ITS EXISTENCE STATUS IN THE LIVER TISSUES AND PERIPHERAL BLOOD LYMPHOCYTES FROM CHRONIC HEPATITIS B PATIENTS

目的探讨慢性乙型肝炎患者外周血淋巴细胞内ＨＢＶ存在状态及其与肝内带毒状况的相关性。方法抽提肝穿刺活检组织、外周血淋巴细胞ＤＮＡ，以ＨＢＶ探针点杂交检测ＨＢＶＤＮＡ，并以Ｓｏｕｔｈｅｒｎ杂交分析其存在状态。结果慢性乙型肝炎患者外周血淋巴细胞、肝组织ＨＢＶＤＮＡ阳性率分别为８７．１％和８３．８％，配对计数的３１例标本χ２检验显示，二者间有一定相关性（ｒ＝０．８１，Ｐ＜０．０５）。整合型与混合型标本比例分别为５５．６％、４４．４％。结论慢性乙型肝炎患者外周血淋巴细胞、肝组织ＨＢＶＤＮＡ有显著相关性，且均有ＨＢＶＤＮＡ整合。

Objective To investigate the existence status of HBV DNA in the peripheral blood lymphocytes (PBL) of chronic HB patients and to study its correlation with the state of HBV infection in the liver.Methods The DNA was extracted from the biopsied liver tissue and PBL of HB patients.The presence of HBV DNA was identified by dot blot using HBV probes and the status of HBV infection in the positive specimens was further determined by Southern blot hybridization.Results The positive rates of HBV DNA were 87.1% and 83.8% in PBL and liver respectively.χ 2 test of the paired data from 31 cases revealed significant correlation in positive rate between PBL and liver (r=0.81,P<0.05).The incidence rates of integrated type and mixed integrated-and-free type were 55.6% and 44.4% respectively.Conclusion The positive rates of HBV DNA in PBL and liver tissue are significantly correlated in chronic HB patients.Integration of HBV DNA into the host′s chromosomes could be seen in all of the cases here studied,which is an event of significance guiding clinicotherapeutical practice.
