养阴清热理气方及其拆方对食管癌裸鼠移植瘤生长的抑制作用

Inhibition of Yangyin Qingre Liqi Formula and Its Constituents on Tumor Growth of Esophageal Carcinoma Transplanted in Nude Mice

目的研究养阴清热理气方及其养阴、清热、理气3个拆方对食管癌裸鼠移植瘤生长的抑制作用。方法将57只BALB/c雌性裸鼠随机分为模型组、全方组、清热组、理气组、养阴组各10只,空白组7只,用人食管癌细胞株EC9706细胞植入裸鼠右胁皮下制造荷瘤裸鼠模型,造模后第2天开始灌胃,各用药组分别给予养阴清热理气方及其拆方,每日1次,共90d。观察各组裸鼠饮食量、体重及移植瘤体积的变化;实验结束后称量各组移植瘤重量并计算抑瘤率。结果各用药组与模型组比较均可增强荷瘤裸鼠精神和活动状态,增加体重(P<0.01),全方组、清热组和养阴组还可提高动物食量(P<0.05)。各用药组对裸鼠皮下移植瘤生长均有不同的抑制作用,瘤体体积、重量与模型组比较差异均有统计学意义(P<0.05或P<0.01)。各组抑瘤率依次为全方组>养阴组>清热组>理气组,全方组、养阴组抑瘤率与清热组、理气组差异有统计学意义(P<0.05)。结论养阴清热理气方及其拆方具有改善荷瘤裸鼠生存状态和抑制肿瘤生长的作用。

Objective To study the effect of Yangyin Qingre Liqi Formula （Formula for nourishing yin, clearing heat and regulating qi） and its 3 constituents on tumor growth of esophageal carcinoma cells transplanted in nude mice. Methods Totally 57 BALB/ c female nude mice were randomized into model group, whole formula group, clearing heat group, regulating qi group and nourishing yin group, 10 mice in each, and blank group of 7 mice. The human esophageal carcinoma EC9706 cells were transplanted in nude mice to make models. On the second day, gastric perfusion was given with the ethanol extract diluent of whole formula and its constituents to nourish yin, clear heat, and regulate qi respectively to the medicinal groups, once per day, totally 90 days. The quantity of food intake, body weight, and tumor size were observed. The tumor weight of all groups was measured after the experiment for calculating the tumor inhibitory rate. Results In all medicinal groups, the general life condition such as spirit and activities of the nude mice was improved and body weight increased, compared with the model group （P〈0. 01）. In whole formula group, clearing heat group and nourishing yin group, the quantity of food intake was also increased （P〈0. 05）. The tumor growth was inhibited in all medicinal groups with a significant difference in tumor size and weight, compared with the model group （P〈0.05 or P〈0. 01）. The tumor inhibitory rate was in an order of whole formula group ~ nourishing yin group 〉clearing heat group 〉 regulating qi group. The difference in tumor inhibitory rate of whole formula group and nourishing yin group was significant （P〈0.05）. Conclusion Yangyin Qingre Liqi Formula and its separated constituents improve the general life conditions and inhibit the growth of esophageal carcinoma in nude mice.
