逆转录PCR检测肝再生过程eNOS的基因表达

摘要 目的探讨eNOS基因表达对肝再生的影响。方法以70%肝切除制备大鼠肝再生模型,注射CCl_4和脾切除引起肝损伤和机体免疫功能低下,FCM检测再生肝DNA的复制,RT-PCR检测eNOS的基因表达,以统计学相关分析判断不同情况下eNOS的改变与再生肝DNA复制的关系。结果肝再生过程伴有eNOS的基因表达,CCl_4肝损害或切除脾脏后肝细胞DNA复制的减少伴有eNOS基因表达的减少;CCl_4肝损害的同时切除脾脏eNOS的基因表达量与单独的CCl_4肝损伤组并无显著性差异。结论 eNOS的基因表达参与了肝再生的调控,其中受到脾脏的影响。但在生理和病理状态下,脾脏的作用方式不同。 Objective To detect the effects of eNOS gene expression on liver regeneration under physiological and pathological conditions. Methods The rat model of liver regeneration were made by partial (70%) hepatectomy. Some rats were injected with CC1_4 and performed to induce liver damage and insufficient im-mune function. The hepatic DNA replication was tested by flow cytometry (FCM), and the eNOS gene expression in liver ceils detected using RT-PCR. The relationship between eNOS change and the hepatic DNA replication was statistically evaluated under different conditions. Results Liver regeneration gener-ally accompanied with the eNOS gene expression. CC1_4-induced liver damage or splenectomy reduced the hepatocyte DNA replication with a decrease of eNOS gene expression. Conclusion The eNOS gene ex-pression might take part in the modulation of liver regeneration. The eNOS gene expression is affected by spleen, and the effects of spleen are different under general or pathological conditions.
