葛根素对人小细胞肺癌H446细胞增殖与凋亡的影响及其作用机制研究 被引量:6

摘要 目的观察葛根素对人小细胞肺癌H446细胞增殖与凋亡的影响,并初步探讨其作用机制。方法采用MTT比色法和流式细胞术检测葛根素对H446细胞增殖抑制和诱导凋亡的作用;免疫组化法检测葛根素对PCNA、Bax和Bcl-2蛋白表达的影响。结果40、80μg/ml浓度的葛根素体外作用24、48h,对H446细胞有促增殖作用,在160~640μg/ml浓度范围内,葛根素能够抑制H446细胞的生长,并呈现良好的剂量依赖和时间依赖关系。435μg/ml的葛根素可以诱导H446细胞凋亡。检测葛根素对PCNA、Bax和Bcl-2蛋白表达的影响显示葛根素能够下调PCNA和Bcl-2蛋白的表达,上调Bax蛋白的表达。结论在一定浓度范围内,葛根素可能通过下调PCNA和Bcl-2蛋白的表达,上调Bax蛋白的表达而诱导H446细胞凋亡。 Objective To explore the effects of puerarin on proliferation and apoptosis in human small cell lung cancer H446 ceils and its possible active mechanisms. Methods The inhibitory effect of puerarin on H446 cells proliferation and apoptosis was determined by MTT assay and by flow cytometry respectively.The expressions levels of PCNA,Bcl-2 and Bax proteins were examined by immunohistochemistry. Results Puerarin could stimulate cell proliferation at a range of concentration of 40-80μg/ml and inhibit proliferation at a range of concentration of 160-640μg/ml with dose and time dependent after treatment for 48h. Puerarin induced cell ap- optosis at concentration of 435 μg/ml . The analysis of protein expression revealed that puerarin significantly reduced expression of PCNA and Bcl-2 and increased Bax. Conclusion Our results demonstrated that puerarin might induce H446 tumor cell apoptisis through down-regulation of PCNA and Bcl-2 protein and up-regulation of Bax protein.
