摘要 目的通过高通量筛选、检测脐血中细胞因子的种类和含量,全面了解脐血浆生物活性成分及表达水平。方法取10位健康足月分娩产妇脐静脉血2 ml和健康成年女性志愿者捐献的外周静脉血2 ml,应用抗体芯片RayBio(?) Biotin Label-basedHuman Antibody ArrayⅠ一次性检测507种细胞因子表达水平,两样本中细胞因子比值大于或小于两倍以上者为有显著差异。结果在蛋白质芯片检测所包括的507种细胞因子中,脐血浆中有94种成分显著高于对照组,28种成分显著低于对照组。结论抗体芯片技术能高通量、平行性检测脐血浆生物活性成分及表达水平,为深入研究脐血细胞因子积累了数据。 Objective To determine the levels of varied cytokines in human umbilical cord blood (HUCB). Methods HUCB was obtained from healthy full-term delivery fetus after the consent of puerperas. Peripheral venous blood (PVB) was collected from healthy voluntary women. As many as 507 kinds of cytokines in plasmas'were simultaneously detected by RayBio Biotin Label-based Human Antibody Array I. When the ratio of a cytokine in HUCB and PVB was 〉 2 or 〈 0. 5, the difference was considered to be remarkable. Results Of the 507 cytokines,94 kinds of cytokines in HUCB increased significantly and 28 kinds in HUCB decreased considerably compared with those in PVB. Conclusions Antibody array may serve as a fast, high-throughput, and sensitive tool for detecting cytokines in HUCB. The relative levels of cytokines obtained in this study provide important information regarding the role of cytokines in HUCB.
