摘要
目的探讨MDM2蛋白、p53在肺腺癌中的表达及其在肺腺癌发生发展中的作用。方法应用免疫组化技术检测41例肺腺癌组织中MDM2、P53的表达。结果MDM2、p53在肺腺癌中的表达率分别为31.71%(13/41),53.66%(22/41);原发灶≥3.0cm者,p53表达率为33.43%±7.28%,高于<3.0cm者6.61%±1.34%,(P<0.01);原发灶≥3.0cm者,MDM2表达率为3.77%±1.13%,低于<3.0cm者24.53%±4.13%(P<0.05)。MDM2表达与突变型P53表达呈负相关性,Sperman等级相关r=-0.54,P<0.05。两者均与肺腺癌病理分型相关,标准回归系数(Beta)值分别为:MDM20.443,p530.74。结论MDM2的表达与P53的表达存在负相关。p53/MDM2存在负反馈机制,是肺腺癌发生发展的分子基础,此外,p53\MDM2各自还存在独立的作用机制。
Objectives To investigate the expression and significance of p53 and MDM2 in lung adenocarcinoma. Methods PCNA、MDM2 and P53 in 41 lung adenocarcinoma specimens were detected by immunohistochenistry. Results The incidence of MDM2 and p53 expression were 31.71% (13/41)and 53.66% (22/41).The expression level of p53 in the huge adenocarcinoma was higher than that of the small adenocarcinoma (33.43%+7.28% vs 6.61%+1.34% ),showing significant difference (P〈0.05). The expression level of MDM2 in the huge adenocareinoma was lower than that of the small adenocareinoma (3.77%+1.13% vs 24.53%+4.13% ), without significant difference(P〈0.05 ). There was a correlation between p53 and MDM2 (r=-0.54,P〈0.05). Conclusion There was a positive correlation between the expression of MDM2 and p53 expression. A negative feedback mechanism exists between p53 and MDM2,it is the molecular basis of the development of lung cancer.
