中电导型钙依赖钾通道在糖尿病大鼠阴茎海绵体中的表达及意义 被引量:3

IKCa in cavernous tissue of rat models of diabetes mellitus

摘要 目的探讨糖尿病对大鼠阴茎海绵体中电导钙激活性钾通道蛋白(IKCa)表达的影响以及意义。方法选用SD雄性大鼠35只,随机选择25只用于制作糖尿病动物模型,剩余10只用于空白对照。造模成功后饲养8周,注射阿朴吗啡(APO),观察大鼠阴茎勃起,同时采用逆转录一聚合酶链反应(RT-PCR)和Westernblot技术检测IKCamRNA和蛋白在大鼠海绵体的表达。结果DM组大鼠阴茎勃起和IKCamRNA及蛋白表达均显著低于STZ组和NDM组(P〈0.05),STZ组和NDM组之间大鼠阴茎勃起情况和IKCamRNA及蛋白表达无统计学意义(P〉0.05)。结论糖尿病可降低大鼠阴茎勃起功能,IKCa在糖尿病大鼠海绵体表达明显降低。糖尿病对大鼠阴茎勃起功能的影响与阴茎海绵体IKCa表达减少密切相关。 Objective To investigate the expression of IKCa in cavernous tissue of rat with diabetes mellitus (DM). Methods All of 35 male Sprague-Dawley rats were divided into 2 groups : control group ( n = 10 ) , DM group ( n = 25 ). After 8 weeks erection function was examined by apomorphine (APO) injection. The expression of IKCa was detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot,respectively. Results Compared with the control group and STZ group, the erection function and expression of IKCa mRNA and protein were significantly decreased in the DM group (P 〈0.05 ,P 〈 0.05 ,P 〈 0.05 ), and there was no significant difference between control group and STZ group (P 〉 0.05 ). Conclusion DM can obviously inhibit IKCa gene and protein expression. The decrease of IKCa gene and protein expression in the cavernous tissue might play a key role in the development of erectile dysfunction in DM rats.
