人骨肉瘤中p53、p21^WAF1/CIP1和增殖细胞核 被引量:1

Expressional studies of p53,p21^WAF1/CIP1 and PCNA in human osteosarcoma

摘要 目的探讨细胞周期相关基因p53、p21^WAF1/CIP1和增殖细胞核抗原(PCNA)在人骨肉瘤中的表达和相互关系。方法应用LSAB法,对40例骨肉瘤、20例骨软骨瘤患者的病变组织和20例正常人骨组织分别以p53、p21^WAF1/CIP1、PCNA蛋白的单克隆抗体作免疫组织化学检测。结果骨肉瘤组织中p53、p21^WAF1/CIP1、PCNA蛋白的阳性反应率和阳性反应强度均明显高于骨软骨瘤和正常骨组织,其差异有统计学意义(P〈0.05)。p53标记指数与p21^WAF1/CIP1标记标数以及p21^WAF1/CIP1与PCNA标记指数间呈正相关(t值分别为2.93和4.20,P〈0.01)。结论骨肉瘤组织中有p53、p21^WAF1/CIP1和PCNA的过表达,其表达的强度与肿瘤的恶性程度和预后相关。 Objective To Study expressional conditions and relationship among cell-cycle-correlated genes p53, p21^WAF1/CIP1 and PCNA. in human osteosarcoma. Methods Positive reaction of monoclonal antibodies anti-p53 protein,anti-p21^WAF1/CIP1 protein and anti-PCNA Protein was observed in 40 ca- ses of osteosareoma (OST) specimens,20 cases of osteochondroma (OCH) and 20 cases of normal osseous tissue (NOT) with LSAB immunohistoehemical technique. Results The positive reaction rate and intensity of p53 ,p21^WAF1/CIP1 and PCNA protein was significantly higher in OST specimens than in OCH and NOT specimens ( P 〈0. 05 ). There was positive correlation in positive staining rate between p53 and p21^WAF1/CIP1 in OST ( r = 0.040 88 ,P 〈 0.01 ). There was also positive correlation in positive staining rate between p21^WAF1/CIP1 and PCNA in OST (r = 0.4262, P 〈 0.01 ). Conclusion There were overexpression of p53,p21^WAF1/CIP1and PCNA in OST. Intensity of the genes positive expression was related to malignant grade of OST.
