摘要
目的:探讨血清对肿瘤细胞凝集的影响,阐明血清中肿瘤细胞凝集因子的生化特性及其在瘤栓形成过程中的作用机制.方法:采用体外细胞凝集实验,通过测定未凝集细胞计算凝集率.结果:正常人、良性瘤患者及恶性实体瘤患者的血清均不同程度诱导高转移潜能人鼻咽癌细胞(CNE-2L2)凝集,细胞凝集率分别为28.2%±8.3%、33.7%±9.1%和68.7%±10.8%.但是层粘连蛋白糖肽处理L2细胞则极大地降低血清凝集活性,而且蛋白酶及高碘酸等处理血清也导致血清凝集活性的极大降低或丧失.然而正常人及良性瘤患者血清对热稳定,恶性瘤患者血清则比较敏感.结论:血清中存在肿瘤细胞凝集因子,其中正常人及良性瘤患者血清中的可能是糖复合物,而恶性瘤患者血清中的则可能是糖复合物和凝集素.它们可能在瘤栓及转移形成过程中起着重要作用,其测定可能有助于恶性肿瘤的临床诊断及预后.
This study was to probe into the effect of human sera on tumor cell aggregation and the biochemical characteristices of tumor cell agglutination factors in human sera, and to elaborate its functional mechanism in the embolication of tumor cells. Methods: The aggregatin of human nasopharyngeal carcinoma cells (CNE - 2L2) with heigh metastatic potential in vitro induced by the sera was quantified by counting the amount of unagglutinated cells. Results: The results showed that the aggregation of CNE - 2L2 cells was induced by the sera of normal persons, benign tumor or malignant tumor patients, and the aggregation incidence of CHE - 212 cells was 28.2% ±8.3% ,33.7±9.1% and 68.7%±?10.8% respectively,but laminin - glycopeptides incubation of CNE - 212 cells extrimely redused the aggregation, moreover, so did the pronase - digestion and periodate - oxidation of sera, however, the agglutinating actively of both normal and benign sera was stable to hot but the activity of malignant sera was more sensitive. Conlusion: These studies demonstrated that there were tumor cell agglutinating factors in human sera, and it is suggested that the factors in normal and benign sera could be glycoconjugates but the onces of malignant sera were probably glycoconjugates and lectin. These factors could potentially play a key role in tumor cell embolization and metastasis. These results maybe provide new important clues and information for clinical diagnosis and prognosis of malignant tumors.
