摘要
本课题包括:①对机会致病寄生虫,包括弓形虫、卡氏肺孢子虫及隐孢子虫动物模型的建立,虫体的分离及提纯;②弓形虫感染的循环抗原(CA)和抗体(IgG、IgM),用竞争抑制酶联免疫吸附试验(IELISA)、金葡菌A蛋白酶联免疫吸附试验(SPAELISA)、直接捕获法酶联免疫吸附试验(DELISA)进行检测;③用地高辛标记探针及聚合酶链反应(PCR)检测核酸,自由引物PCR鉴别弓形虫株以及生物特性;④用流式细胞仪对卡氏肺孢子虫及弓形虫的DNA、RNA及蛋白质含量的动力学进行了分析;⑤用激光微束系统对含8个子孢子成熟包囊施行手术,切除4个或2个子孢子后,包囊仍具有感染力。
The contents of serial studies involve isolation,purification and animal experimental model built up of opportunistic pathogenetic parasitic protozoan including Pnemocystis,Cryptosporidium and Toxoplasma; detection of circulating antigen (CA) and antibody (IgG,IgM) by I ELISA,SPA ELISA,D ELISA; determination of DNA by PCR and digoxigenin labeled probe; identification of Toxoplasma strain by RP PCR and biological characters. Dynamics of Pneumocystis and Toxoplasma DNA,RNA and protein contents were analysed by Laser Flow Cytometry. Microsurgery on P. carinii was made with ultrashort pulse laser microbeam system. Cysts being cut off 4 or 2 intrabodies had the ability to re infect made. It suggested that cyst with 8 intrabodies was mature one, etc.
