摘要
目的:建立赤芝RP-HPLC色谱特征图谱,为赤芝药材的质量控制提供依据。方法:以灵芝酸A为内参照物,采用Kro-masil C18(250 mm×4.6 mm,5μm)色谱柱,以乙腈(A)-0.03%磷酸水溶液(B)为流动相,进行梯度洗脱(0~35 min,32%→33%A;35~38 min,33%→37%A;38~60 min,37%→40%A;60~70 min,40%A),流速1.0 mL.min-1,检测波长252 nm,柱温35℃。结果:获得了较为理想的赤芝药材HPLC特征图谱,确定了19个共有峰。根据聚类分析结果,将赤芝药材分为2类。结论:该方法简便、准确、可靠,可用于赤芝药材的质量控制。
objective: To establish the HPLC specific chromatogram of Ganoderma lucidum. Methods: 16 samples of G.lucidum from different producing areas were determined by SHIMADZU LC-10AD DAD-HPLC under the chromatographic conditions: column by Kromasil C18(250 mm×4.6 mm,5 μm),acetonitrile(A)-0.03%phosphoric acid solution(B) with gradient elution(0-35 min,32%→33%A;35-38 min,33%→37%A;38-60 min,37%→40%A;60-70 min,40%A) at a flow rate of 1.0 mL·min^-1,wavelength 252 nm,column temperature 35 ℃.Results:A satisfactory method for HPLC specific chromatogram determination of G.lucidum was established,19 characteristic peaks in the HPLC specific chromatogram were indicated.The 16 batches of G.lucidum were recommended into two based on the results of cluster analysis. Conclusion:The method is simple and accuracy with good reproducibility and can be used as quality control method for G. lucidum.
