GC-MS analysis of chemical constituents of the essential oil from Plumula nelumbinis by different extraction methods

摘要 目的:分析比较采用超临界CO2流体萃取法与水蒸气蒸馏法提取的莲子心提取物中挥发性化学成分的异同。方法:使用水蒸馏提取法和超临界CO2萃取技术从莲子心中提取挥发性成分,用归一化法测定其百分含量。用气相色谱-质谱(GC-MS)计算机联用技术分离鉴定其中的化学组成。结果:莲子心超临界CO2流体萃取物中初步鉴定了25种成分,主要成分为:2-氯亚油酸乙酯(42.85%)、n-十六烷酸(21.06%)、Z-9,17-十八碳二烯醛(11.99%);水蒸气蒸馏法提取挥发油初步鉴定了36种成分,主要成分为14-甲基-十五烷酸甲酯(11.43%)、8,11-十八碳二烯酸甲酯(8.95%)、十五烷(6.89%)。结论:超临界CO2流体萃取法提取的挥发油能更真实、全面地反映药材中的化学成分。 Objective:To compare the chemical constituents of the essential oil from Plumula nelumbinis by supercritical fluid extraction and steam distillation by using the GC-MS Analysis. Methods: The essential oils were extracted from the plumule of Nelumbo nucifera Gaertn. by supercritical fluid extraction (SFE) and steam distillation (SD). The amounts of the components from the essential oil were determined by normalization method. The components were separated and identified by GC-MS. Chromatographic conditions were: Capillary column HP-5MS 5% Phenyl Methyl Siloxane Capillary 30.0m×250μm×0.25μm nominal; Injector temperature 250℃;Program temperature: initial column temperature 50℃,maintained 3 min, and increased to 250℃ at the rate of 4℃·min^-1,held 7 min; Splitless injection. Result: There were 25 components from Plumula nelumbinis by SFE which separated and identified by GC-MS. The major constituents are n-Hexadecanoicacid(21.06%),2-Chloroethyllinoleate(42.85%) and (Z)-,9,17-Octadecadienal (11.99%). There were 36 components from Plumula nelumbinis by SD. The major constituents are Pentadecanoic acid, 14-methyl-, methyl ester (11.43%),8,11-Octadecadienoic acid, methyl ester (8.95%) and Pentadecane(6.89%). Conclusion: The result got by SFE method can reflect the chemical components of the medicinal materials more accurately and more comprehensively.
