海风藤抑制淀粉样前体蛋白基因表达的研究

Inhibition of β-amyloid Precursor Protein Gene Expression by Haifengteng

目的：研究海风藤对人类神经母细胞瘤细胞系列淀粉样前体蛋白（βＡＰＰ）基因表达的抑制作用。方法：应用Ｎｏｒｔｈｅｒｎ印迹杂交观察βＡＰＰｍＲＮＡ的变化，应用细胞存活率、乳酸脱氢酶（ＬＤＨ）释放、细胞集落观察海风藤对神经细胞的直接作用。结果：海风藤选择性的抑制βＡＰＰ基因表达。实验浓度内未发现细胞毒性作用。结论：βＡＰＰ与Ａｌｚｈｅｉｍｅｒ’ｓ病（ＡＤ）的发病关系密切。

Objective:To study the inhibition of β amyloid precursor protein(β APP) gene expression by Haifengteng.Method:Northern Blot analysis was used to investigate the level of β APP mRNA,and cell viability,LDH release and colony formation assay were used to study the effect of Hairengteng of neurons.Result:Haifengteng inhibited the expression of β APP gene selectively without causing cell injuries.Conclusion:β APP is closely related to Alzheimer's disease.The findings in this study could be a basis for Haifengteng to have thrapeutic potentials in treating Alzheimer's disease.
