摘要
目的探究转化生长因子-β(TGF-β)在移植后淋巴组织增生性疾病(PTLD)发生、发展中的作用。方法建立人鼠PTLD嵌合模型40个,随机分为观察组及对照组各20个,观察组注射抗TGF-β单克隆抗体125μg/只,每周3次,对照组注射等量PBS缓冲液至小鼠死亡。注射第9周每组各取10只测定两组成瘤率并通过流式细胞仪来检测肿瘤中细胞免疫表型;比较每组另10只的生存时间。结果观察组成瘤率低于对照组(P=0.003,<0.01),而生存时间明显高于对照组(t=35.78,P<0.05),两组T、B淋巴细胞水平有统计学意义(P<0.05)。结论TGF-β可以促进PTLD的发生。
Objective To investigate the role of the transforming growth factor-13 (TGF-β) in the occurrence and development of post-transplant lymphoproliferative disease (PTLD). Methods Forty human mouse chimeric models of PTLD were established and randomly divided into observed group and control group with 20 in each. The observed group was given intraperitoneal injection of anti-TGF-β monoelonal antibody 125 μg/once, 3 times per week, and the eontrol group was given equivalent PBS buffer. The formation rate of tumor in 20 mice of the two groups was determinated 9 weeks later , and the survival time of another 20 mice was compared. Results The formation rate of tumor and the survival time in the observed group were significantly higher than those in the control group ( t = 35.78, P 〈 0.05 ). There was statistical significance of T, B lymphocytes level in two groups( P 〈 0.05 ). Conclusion TGF-β can promote the occurrence of PTLD.
