摘要
对根霉3.010所产纤维素酶酶学特性进行了分析研究。实验结果显示:根霉在培养的第6d~8d为纤维素酶旺盛分泌期,其最适反应温度为50℃左右,且在40℃以下热稳定性较好;纤维素酶的最适反应pH值为4.0~6.0,在pH4.0~7.0时稳定性较好。在1.0%的浓度下,CaCl_2、MnSO_4和CoCl_2均对该酶有激活作用:而KCl、ZnSO_4和CuSO_4则对该酶活力有不同程度的抑制作用;NaCl、FeSO_4和Mg_2SO_4对根霉纤维素酶活力影响不明显。
The enzymatic properties of cellulases produced by Rhizopus 3.010 were studied in this paper. The results showed that yield of cellulose by Rhizopus 3.010 reached the highest after 6-8 d inoculation. The optimum temperature for cellulase production was 50℃ and stability of enzyme was better when temperature was lower than 40℃. The optimal pH of the enzyme was 4.0-6.0 and it has a better stability when pH in the range of 4.0-7.0. Presence of 1.0% CaCl2, 1.0% MnSO4 and 1.0% COCl2 could improve cellulases activity, while 1.0% KCl, 1.0% ZnSO4 and 1.0% CuSO4 may inhibit cellulases activity. Presence of 1.0% NaCl, 1,0% FeSO4 and 1.0% Mg2SO4 has no significant effect on the cellulases activity.
