摘要
将制首乌及其 4种中成药在酸性条件下水解 ,提取 ,然后以C18为固定相 ,甲醇— 0 2mol/L磷酸二氢钠溶液 (80∶2 0 ,磷酸调节pH 3 0 )为流动相 ,2 89nm为检测波长 ,测定其中大黄素含量 ,建立了测定制首乌及其中成药中大黄素总量的反相高效液相色谱法。同时对样品的水解及提取条件进行了详细考察。在所拟条件下 ,大黄素线性范围为 32 8~ 4 10ng ,平均加样回收率为 97 83%。
A RP-HPLC method has been developed to determine the emodin in Radix Polygoni Multiflori Preparata and four kinds of related Chinese patent medicines containing Radix Polygoni Multiflori Preparata,using a Shimpack CLC-ODS column with a mobile phase of methanol-0 2 mol/L NaH 2PO 4(80∶20,adjust to pH 3 0 with H 3PO 4)at a flow rate of 1 mL/min and a UV detector at 289 nm.After hydrolysized in acidic medium,the total emodin of the samples are extracted from the hydrolysates with ether and determined by the RP-HPLC method.The linear range of emodin is 32 8~410 ng,the mean recovery is 97 83 %.
