摘要
[目的]探讨高温不同处理对MS培养基pH值的影响。[方法]测定培养基经高温高压与普通高温煮沸处理前后的pH值,并进行比较分析。[结果]2种高温方式单独处理后分别使25 ml的培养基pH值下降了0.52与0.57,培养基经过先普通高温煮沸后高温高压处理的pH值下降幅度大于先高温高压后普通高温煮沸处理的pH值下降幅度。[结论]高温不同处理均能引起pH值的下降,下降的幅度依具体处理而异。
[ Objective ] The research aimed to investigate the effects of different high temperature treatments on pH value of MS culture medium. [ Method] The pH value of Culture medium was determined and compared before and after treatment with high temperature and high pressure and the ordinary boiling temperature. [ Result] pH value of 25 ml culture medium dropped 0.52 and 0.57 after two separate high temperature treatment respectively. The decreased amplitude of pH value in the medium after treatment with the ordinary boiling temperature and then high temperature and high pressure was greater than that with high temperature and high pressure and then the ordinary boiling temperature. [ Conclusion ] Different high temperature treatments can decrease the pH value, but the decrease amplitude is different according to concrete treatments.
