搜风祛痰中药对AngⅡ诱导人脐静脉内皮细胞LOX-1 mRNA及NF-κB mRNA表达的影响 被引量:4

摘要 目的观察搜风祛痰中药对AngⅡ诱导体外培养人脐静脉内皮细胞凝素样氧化低密度脂蛋白受体1(LOX-1)与核因子-κB基因表达的影响。方法制备家兔中药复方活心汤含药血清。采用酶消化法体外培养HUVECs2~3代用于实验。采用RT-PCR法测定脐静脉血管内皮细胞LOX-1 mRNA水平。随机分为正常对照组、AngⅡ组、AngⅡ+低剂量中药血清组、AngⅡ+中剂量中药血清组、AngⅡ+高剂量中药血清组。用中药先孵育2h后再加入0.5μmol/LAngⅡ刺激2h,取出细胞盖玻片固定。结果与对照组比较,活心汤含药血清可抑制AngⅡ导致的细胞损伤,减少LOX-1mRNA及NF-κB mRNA的表达。结论活心汤可抑制AngⅡ所致的HUVECs损伤,保护血管内皮细胞功能。 Objective: To observe the effect of expelling wind and expectorant Chinese medicine on the expression of LOX-1 mRNA and NF-κB mRNA induced by angiotensin Ⅱ(Ang Ⅱ) in human umbilical vein endothelial cells (HUVECs) . Methods: The serum containing metabolic ingredients of Huoxin Decoction was taken from rabbits. HUVECs 2-3 were cultured in vitro by enzyme digestion. LOX-1 mRNA was determined by RT-PCR. HUVECs were randomly divided into five groups: normal control group, Ang Ⅱ group, Ang Ⅱ low dosage drug serum group, Ang Ⅱ middle dosage drug serum group and Ang Ⅱ high dosage drug serum group. HUVECs were preincubated by Chinese medicine for 2h,and then were added Ang Ⅱ 0.5μmol/L to stimulate for 2h. At last,the cells were taken out to fix by coverslip. Results: Compared with control group, Huoxin Decoction drug serum could inhibit the injury of endothelial cells and decrease the expression of LOX-1 mRNA and NF-κB mRNA. Conclusion: Huoxin Decoction can inhibit the injury of HUVECs induced by Ang Ⅱ and protect vascular endothelial cells function.
