摘要
目的研究Semaphorin3A(SEMA3A)及其受体Neuropilin1(NRP1)和PlexinA1在口腔癌组织和细胞(ACC2,ACCM和TCa)中的表达情况,并推测其在口腔癌生成中的可能意义。方法应用RT-PCR和Western bloting以及免疫组织化学方法检测SEMA3A、NRP-1及Plexin A1在口腔癌(ACC2,ACCM和TCa)组织和细胞中的表达情况。结果在所有口腔癌组织和癌细胞中均可检测到NRP-1和Plexin A1的表达,但SEMA3A相对表达较弱。Western bloting、RT-PCR提示NRP-1,Plexin A1呈高表达,而SEMA3A表达较低。在免疫组织化学中显示癌及癌周组织中NRP-1均呈高表达,且癌巢中的表达明显高于癌间隙组织,SEMA3A呈较弱表达。结论三种不同头颈肿瘤及细胞系中NRP-1,Plexin A1表达均较高,提示在肿瘤的发展进程中,Plexin A1可能对肿瘤本身的发生,增殖及肿瘤的血管生成均可能起到了重要的作用,这种作用通过其它的Semaphorin家族成员实现。
Objective To investigate the expression and distribution of the semphorin 3A (SEMA 3A) and its receptor neuropilin-1 and Plexin A1 in the tissue and cells of the head and neck carcinoma(ACC2,ACCM and TCa) and their potential function in the tumor.Methods Reverse transcription polymerase chain reaction (RT-PCR) and the western bloting were used to survey the expression of sema3F,neuropilin-1 and Plexin A1 with specific primers and antibody.Immunohistochemistry staining for SEMA 3A,neuropilin-1 and Plexin A1 with specific primers and antibody. Immunohistochemistry stalning for SEMA 3A, neuropilin1 and Plexin AI were also performed in the tissues. Results Neuropilin-I ,Plexin AI mRNA were detected in all the oral carcinoma tissue and the cells,but very weak signals for SEMA 3A were detected. Western bloting indicated the high expression of the NRP-1, Plexin AI ,and the low expression of the SEMA 3A. Immunohistochemistry indicated that NRP1 were highly expressed in the tissue of the carcinoma,especially in the cancer nest. Conclusions The high expression of Plexin A1 indicated that Plexin A1 may play an importmant part in the process of the tumour generation, both in the tumor proliferation and in the angiogenesis, and this role may combine with other semaphorins.
