氟对体外培养人牙胚TGF-β1表达影响的实验研究 被引量:2

摘要 目的研究氟对牙胚发育早期TGF-β1表达的影响。方法应用牙胚体外培养模型,观察低剂量(20mg/L)和中剂量(50mg/L)的氟作用于分泌前期牙胚后,TGF-β1表达情况的变化。采用图像分析仪对免疫组化染色结果进行灰度分析,并对灰度值进行统计学检验。结果在分泌前期TGF-β1的表达主要在造釉器,在20mg/L、50mg/L的氟条件下从培养第4天开始TGF-β1的表达显著降低。结论在牙胚发育早期,TGF-β1主要在上皮表达,TGF-β1可能通过旁分泌和自分泌来调节内釉上皮细胞、牙乳头细胞的分化及基质的合成与分泌,低剂量和中剂量的氟显著抑制TGF-β1的表达,提示氟可能通过抑制TGF-β1的表达而抑制了内釉上皮细胞和牙乳头细胞的分化及随后的基质合成与分泌。 Objective To observe the influence of fluoride on the expression of TGF-β1 (transforming growth factorβ1)in the tooth germ cultured in vitro during presecretory stage.Methods We cultured the human tooth germ during presecretory stage and observed the expression of TGF-β1 in the condition of low dosage(20 mg/L) and medium dosage(50 mg/L) fluoride in the culture medium.The sections were undertaken with the immunohistochemistry staining.Then the gray scale analysis was done with the image analysing system.The results were analyzed with statistical methods. Results The expression of TGF-β1 was found mostly in the enamel organ during presecretory stage. The expression degraded significantly from the 4th day in the condition of 20 mg/L and 50 mg/L fluoride. Conclusions During the presecretory stage,the expression of TGF-β1 was found mostly in the epithelium. TGF-β1 possibly adjusted the differentiation of the inner enamal epithelium cell and dental papilla cell,the matrix synthesis and secretion sequently by paracrine and autocrine. The expression was inhibited significantly in degrading the expression of TGF-β1 to inhibit the differentiation of the inner enamal the condition of low dosage and medium dosage fluoride. It was suggested that fluoride degraded the expression of TGF-β1 to inhibit the differentiation of the inner enamal epithelium cell and dental papilla cell,the matrix synthesis and secretion sequently.
