HPLC测定青阳参中的青阳参苷元

Determination of cynanchogenin in Cynanchum otophyllum Scheid by HPLC

目的建立测定青阳参中青阳参苷元含量的方法。方法采用HPLC法,色谱柱为Hypersil C18柱(150mm×4.6mm,5μm),流动相为甲醇-0.4%磷酸溶液(1:1),流速为0.8mL·min-1,柱温为35℃,检测波长为259nm。结果青阳参苷元的质量浓度1.0～192.8μg·mL-1与峰面积呈良好的线性关系(r=0.9999,n=6),平均回收率为99.0%(n=9)。结论所用方法简单、快速、准确,可作为青阳参的质量控制方法。

OBJECTIVE To estabish an HPLC method for determination of cynanchogenin in Cynanchum otophyUum Scheid. METHODS Hypersil C18 column was used. The mobile phase consisted of CHsOH -0.4% H3PO4 （ 1 ： 1 ）. The flow rate was 0.8 mL·min^-1. The temperature of column was 35℃ and the detection wavelength was set at 259 nm. RESULTS The linear range for cynanchogenin was 1.0 - 192.8 μg·mL ^- 1 （ r = 0. 9999 ）. The average recovery was 99.0% with RSD of 2.2 %. CONCLUSION The method is simple, rapid, accurate and can be used to determine cynanchogenin in Cynanchum otophyllum Scheid.
