牡丹鳞芽诱导与增殖过程中影响因子研究 被引量:5

摘要 以牡丹鳞芽为外植体,研究不同品种类型、基本培养基种类、取材时间、暗处理、植物生长调节物质种类及其质量浓度、继代周期等因素对鳞芽诱导与增殖的影响.结果表明,影响鳞芽诱导的主要因子是品种和采集时间,鳞芽适宜的采集时间为01~02月;不同GA3质量浓度、暗处理和基本培养基对鳞芽诱导的影响较小;牡丹试管苗增殖中,4种基本培养基类型相比较,MS+Ca^2+效果明显;培养基中添加6-BA和IAA对试管苗增殖率有一定影响,最佳培养基组合为:MS+Ca^2++6-BA 2.0 mg.L^-1+IAA 0.3 mg.L^-1,增殖培养适宜的继代周期为35 d. The axillary buds of Paeonia suffruticosa were selected as explants to study the effects of cultivars,basic mediums,time of axillary buds collection,dark treatment,medium types and mass concentration of plant growth regulators and subculture cycle on induction and multiplication of Paeonia suffruticosa axillary bud in vitro.The results showed that cultivars and collection time of axillary buds were the main factors affecting its induction,January and February was the suitable collection time, but GA3 mass concentrations, dark treatment and basic mediums had less influence on axillary buds induction. In the four basic mediums, the medium with MS + Ca^2+ was the best one for plantlet multiplication, the medium with 6 - BA and IAA had some influence on plantlet multiplication coefficient, the best multiplication mediums was MS + Ca^2+ + 6 - BA 2.0 mg · L^- 1 + IAA 0.3 mg · L^- 1, and the suitable subculture cycle was 35 d for plantlet multiplication.
