摘要
以小麦稳定表达的肌动蛋白(Actin)为对照,利用半定量RT-PCR技术对小麦禾谷胞囊线虫(Heterodera ave-nae)接种后在不同抗性品种不同时期不同部位的几丁质酶基因Cht4的表达量进行了分析.结果表明,接种12 h后,Cht4的表达量无明显的增加,抗、感品种之间差异不明显;接种36 h后,抗、感品种中的Cht4的表达量都明显增加,抗性品种中的表达量明显高于感病品种,叶部表达量高于根部的表达量;接种60 h后,抗、感品种中Cht4的表达量都维持在较高的表达水平,抗病品种比感病品种中Cht4的表达量高;接种84 h后,感病品种与接种前相比变化不明显,而抗病品种的表达量仍高于接种前的表达量.
Expression analysis about wheat chitinase gene,in the different growing stages and different positions of different resistance cultivars after CCN inoculation,was carried by semi-Q RT-PCR,using wheat Actin gene as a control.The result showed that the quantity of chitinase gene expression obvious increased after 12 h inoculation,and there were no obvious differences between resistant and susceptible cultivars.After 36 h inoculation,the expression quantity in both cultivars increased obviously,and the quantity in resistant cuhivars was more than susceptible cultivars, and in leaves was more than in roots. After 60 h inoculation, the gene expression quantity in resistant and susceptible cuhivars was keeping a high level, and the former was more than the latter. The expression in susceptible cultivars after 84 h inoculation didn' t have obvious difference from the ones without inoculation, but the expres- sion quantity in resistant cuhivars was still higher than those without inoculation.
