摘要
以传统的Folin-酚方法为基础,分别采用BSA,Tryptone和Peptone作为标准物质,制作小肽质量浓度测定用标准曲线。结合分子筛Superdex 30对Tryptone和Peptone进行分析,确定了Peptone为最佳标准物,以其制作的Peptone-folin-酚标准曲线,相关系数R=0.99936,与小肽质量浓度的相关性良好。在此基础上采用建立的标准曲线测定各种发酵条件的小肽质量浓度。结果表明,用Lacto-bacillus delbrueckii ssp.bulgaricus LB340菌株发酵的牛乳产生的小肽质量浓度最高,为乳源性活性肽的制备和研究奠定基础。
In this study, we investigated the method to determine peptides concentration. Based on traditional Folin-phenol method, we established concentration standard curves using BSA, Tryptone and Peptone as standard samples, respectively. Then the gel chromatography Superdex 30 was used to analyze the component ratio of the peptides in Tryptone and Peptone. In conclusion, we selected Peptone as the best standard sample, and the correlation coefficient (R) of the standard curve is 0.99936. With developed Peptone- folin-phenol method, we tested the peptides concentration in fermented milk with different strains and conditions. The result showed that peptides could be produced with the highest concentration from milk protein using Lactobacillus delbrueckii ssp. bulgaricus LB340
