摘要 通过单因素试验和L27(313)正交试验,确定了乳酸菌富硒的最佳发酵条件。正交试验结果表明,影响乳酸菌总硒量的主要因素是培养温度、接种量、亚硒酸钠浓度和培养温度与接种量的一次交互作用。最佳发酵条件为培养基初始pH为6.6,亚硒酸钠浓度为18μg/mL,装液量为150 mL/250 mL三角瓶,接种量5.5%,培养温度42℃,培养时间45.5 h。在此优化条件下,富硒乳酸菌生物量可达6.85 g/L,细胞硒含量达1100μg/g,硒总含量可达7536μg/L,细胞硒含量的92.15%为有机硒。 A optimized fermentation condition of Se--enriched Lactobacillus was confirmed by single factor tests and orthogonal test L27 (3^13). The results of orthogonal test showed that the main influence factors were culture temperature, inoculating content, Na2SeOa concentration, linear interaction of culture temperature and inoculating content on Se-enriched content of Lactobacillus. The fermentation condition was optimized as: the initial pH of culture medium was 6.6 and Na2SeO3 concentration was 18 μg/mL,The inoculating content was about 5.5% , 150 mL culture in 250 mL flasks, the temperature was at 42℃ and the culture time was 45.5 h. Under the optimized conditions, the biomass(dry weight) reached to 6.85 g/L with the Se content of the cells reached to 1100 μg/g, and the total Se content reached to 7536 μg/L, in which the organic Se is up to 92.15%.
