摘要
采用水浴法、微波辅助提取法与超声-微波协同萃取法提取黑果枸杞叶片多糖,确定最适提取方法。通过单因素试验和正交试验确定最优提取工艺。采用蒽酮-硫酸法测定多糖含量。结果表明:最佳提取方法为超声-微波协同萃取法。最优提取条件是:料液比为1:25,提取温度为90℃,提取时间为30min,多糖平均提取率为10.07%,RSD=1.60%(n=3)。回收率100.7%,RSD=1.84%(n=3)。
Water-bath, microwave assisted and ultrasonic-microwave assisted extraction teehniques were applied to extract polysaceharides from Lyeium rethenieum leaves and the optimal extraction method was selected by compared with others. Extraction Conditions were assured by a single factor and orthogonal experiments. Content of polysaeeharides was determined by anthrone-sulphuric acid colorimetry. The results showed that Ultrasonicmicrowave extraction was the optimum extraction method. The results of an orthogonal experiment indieated that the effectiveness factors on polysaeeharides content was the order as follows: temperature 〉 extraction time 〉 solid-liquid ratio. The technological parameters were as follows: solid-liquid ratio was 1:25, extraction temperature was 90 ℃, extraction time was 30 rain. The extraction rate of polysaeeharides was 10.07 %,RSD = 1.60 % (n =3 ). Recovery rate was 100.7 %, RSD=1.84 %(n=3).
